Scientists Find Brain Mechanism Behind Age-Related Memory Loss
July 1, 2022 – Scientists at Johns Hopkins University have identified a mechanism in the brain behind age-related memory loss, expanding our knowledge of the inner workings of the aging brain and possibly opening the door to new Alzheimer’s treatments.
The researchers looked at the hippocampus, a part of the brain thought to store long-term memories.
Neurons there are responsible for a pair of memory functions – called pattern separation and pattern completion – that work together in young, healthy brains. These functions can swing out of balance with age, impacting memory.
The Johns Hopkins team may have discovered what causes this imbalance. Their findings – reported in a new paper in the journal Current Biology – may not only help us improve dementia treatments, but even prevent or delay a loss of thinking skills in the first place, the researchers say. Pattern Separation vs. Pattern Completion To understand how the hippocampus changes with age, the researchers looked at rats’ brains. In rats and in humans, pattern separation and pattern completion are present, controlled by neurons in the hippocampus. As the name suggests, pattern completion is when you take a few details or fragments of information – a few notes of music, or the start of a famous movie quote – and your brain retrieves the full memory. Pattern separation, on the other hand, is being able to tell similar observations or experiences apart (like two visits to the same restaurant) to be stored as separate memories.
These functions occur along a gradient across a tiny region called CA3. That gradient, the study found, disappears with aging, says lead study author Hey-Kyoung Lee, PhD, an assistant research scientist at the university’s Zanvyl Krieger Mind/Brain Institute. “The main consequence of the loss,” Lee says, “is that pattern completion becomes more dominant in rats as they age.” What’s Happening in the Brain Neurons responsible for pattern completion occupy the “distal” end of CA3, while those in charge of pattern separation reside at the “proximal” end. Lee says prior studies had not examined the proximal and distal regions separately, as she and her team did in this study. What was surprising, says Lee, “was that hyperactivity in aging was observed toward the proximal CA3 region, not the expected distal region.” Contrary to their expectations, that hyperactivity did not enhance function in that area but rather dampened it. Hence: “There is diminished pattern separation and augmented pattern completion,” says Lee.
As pattern completion dominates, pattern separation fades, Lee says. This may make it harder for older adults to separate memories – they may recall a certain restaurant they’d been to but not be able to separate what happened during one visit versus another. Why Do Some Older Adults Stay Sharp? But that memory impairment does not happen to everyone, and it doesn’t happen to all health rats either. In fact, the researchers found that some older rats performed spatial-learning tasks as well as young rats did – even though their brains were already beginning to favor pattern completion.
If we can better understand why this happens, we may uncover new therapies for age-related memory loss, Lee says. Co-author Michela Gallagher’s team previously demonstrated that the anti-epilepsy drug levetiracetam improves memory performance by reducing hyperactivity in the hippocampus.
The extra detail this study adds may allow scientists to better aim such drugs in the future, Lee speculates. “It would give us better control of where we could possibly target the deficits we see.”

Ricky Martin Reportedly Hit with DV Restraining Order in Puerto Rico

We are very confident that when the true facts come out in this matter our client Ricky Martin will be fully vindicated.” Ricky Martin has been accused of domestic violence in Puerto Rico — although it appears the alleged victim may be someone other than his spouse … this per several reports. Multiple Spanish outlets that operate in PR — including Telemundo — reported Saturday that a protective order is in the process of being filed and executed against the pop star in a municipality near San Juan … citing law enforcement officials who confirmed the complaint falls within their Law 54 statute, which deals with DV. The reports state an anonymous person — who goes unnamed — recently appeared before a judge in their own right to seek the TRO without going to the police first and without Ricky being present … and the order was apparently granted based on evidence presented. The purported order goes on to reportedly say this … “the petitioning party demonstrated that there is a substantial possibility of immediate risk of abuse.” As a result of the order reportedly being granted, Ricky has to steer clear of this person — including avoiding all communication — until this can get cleared up before a judge. As of Saturday, cops travel in Puerto Rico told AP they had not located Ricky to serve him. Of course, Ricky has several residences … including property in Puerto Rico. It’s interesting this is surfacing now — days after he was sued by his ex-manager, who claimed to have knowledge of a “career-ending allegation” from which she says she helped shield him. Taron Egerton Hopes He’ll Get to Play Wolverine in the MCU
Image: 20th Century Studios Ever since Fox got themselves snatched up Disney and Kevin Feige made it clear that the mutants would be making their way to the MCU eventually, there’s been no shortage of speculation about who’ll be playing the new batch of X-Men. Moreso than Professor X or Magneto, Wolverine is the one that everyone’s got some opinion on. These days, we now know there’s nothing stopping Hugh Jackman from coming back to the role (and the frankly absurd diet needed to maintain it) that launched him into cinematic stardom. But there’s been no shortage of folks who want someone new to take the reigns, and one actor is now specifically making their desires clear rather than simply dancing around the subject and hoping to be John Krasinski’d into an eventual film.
Specifically, it’s Taron Egerton of Kingsman and Rocketman fame (or more importantly, Moomintroll ) who wants to get in on the comic book action. He said as such to the New York Times; while promoting his upcoming Apple TV+ show Black Bird, Egerton admitted that he has spoken with Marvel Studios employees, including Kevin Feige, about getting involved in the superhero game. While it’s something he’s hoping to take a shot at, he also knows that there’s some big shoes to fill, regardless of whoever eventually plays the character. “I’d be excited, but I’d be apprehensive as well,” he said. “Hugh is so associated with the role that I’d wonder if it’d be very difficult for someone else to do it.” Back in 2019, the actor talked about being health aware of fans wanting him in the role, and was flattered that they were rooting for him. Talking with Feige raises the chances of Egerton getting a shot to play Wolverine just slightly, but it’s equally possible that Feige and crew have someone else in mind for him to play. Since Marvel has just so much stuff in various stages of production, we won’t really know what Egerton’s future is until we know. Maybe he’ll wind up being Wolverine, mayhaps he’ll get locked in for Human Torch or Nova . (Maybe he’d be a good Cyclops?) Either way, it’s doubtful that he won’t wind up playing a superhero eventually, since basically everyone does…it’s just a matter of whom it’ll be.
